Summary:Various polysaccharide components in aloe vera, including acemannan, have activity as an anti-wrinkle. Acemannan has a high molecular weight and hydrophilic thus it has limited penetration through the skin. This research aims to develop formulation of acemannan nanoemulsion having better penetration through the skin. Acemannan nanoemulsion consisted of coconut oil, Tween 80 and propylene glycol as the oil phase, surfactant and cosurfactant, prespectively. Acemannan was dispersed into the oil phase and stabilized by Tween 20 prior to emulsification. Acemannan nanoemulsion was evaluated by organoleptic, viscosity, pH, globule size, and polydispersity index. Skin penetration of optimum formula was tested in vitro using Franz diffusion cells. Acemannan dispersion in coconut oil with a clear appearance was obtained by co-lyophilization. The optimum formula of acemannan nanoemulsion consisted of 15.4% coconut oil, 30% Tween 80, 10% propylene glycol, 1.35% tween 20 and 0.5% acemannan. Incorporation acemannan in nanoemulsion base increased significantly the globule size from 19.33 ± 3.23 nm to 195.55 ± 7.85 nm (p <0.05) however didn????t change the viscosity (103.5 ± 0.408 cps), pH (4, 63), and polydispersity index (0.325 ± 0.045). In the penetration study, nanoemulsion that contained acemannan solid-in-coconut oil lyophilizate could improved skin compered to nanoemulsion containing acemannan without co-lyophilization (p<0.05). acemannan nanoemulsion optimum formula with good penetration has the potential to be developed into an anti-aging product.
