Extract of mangosteen’s pericarp has many pharmacological effects such as antioxidant,
anticancer, antidiabetic, antiinflamation, antifungi and immunomodulator. Acne or acne
vulgaris is a disease which cause pilocebaceus folicle clogging. Propionibacterium acnes is
one of bacteria that can cause acne. The aim of this study is to formulate pericarp extract of
mangosteen in microemulsion that can be used for topical as antiacne and in vitro
antibacterial test againts P. acnes. Crude drug was extracted by ethanol 96% using reflux
method. Extract was concentrated with rotavapor and then, phytochemical screening and
characterization were performed. Microemulsion were optimized based on its total sufactantcosurfactant and oil used, ratio of surfactan-cosurfactant, optimum extract concentration that
can be used in formulation. The result of optimization was used for final microemulsion
formulation. the concentrated extract was formulated into microemulsion. Formulated
extract was evaluated based on physical aspect of dosage form and in vitro activity study
using microdilution test for determining minimum inhibition concentration (MIC) and disc
diffusion test was also performed. Bacterial inhibition of microemulsion was compared with
non-formulated extract to observe the activity after formulation. Mangosteen pericarp extract
contained flavonoid, tannin, saponin, and quinone. Density of extract was 2,02 g/mL, water
extractable matter 26.20%, ethanol extractable matter 66.03%. Polisorbate 80 and gliserin
were used as surfactan and cosurfactan. The formula consisted of virgin coconut oil 5%,
Extract 1 %, total surfactan 40% with ratio of surfactan and cosurfactan 2:1 and water 54%.
Extract-loaded microemulsion had similar antimicrobial inhibitory compared to the nonformulated extract and had minimum inhibition concentration (MIC) at 312.5 µg/mL.
