The use of antibiotic for therapy is based of some considerations like : its selectivity to infectious microorganism and its effectivity to destroy that microorganism, and to have the least potential to create toxicity of allergical reaction or another risks for the patients. Penicillin is a group of antibiotic which is used in common as it fulfils those criteria. The enzyme which plays a role in hydrolisis of benzilpenicillin to produce 6-amino penicilic acid (6-APA ), an essential material of semisynthetic antibiotic derived from penicillin, is penicillin acylase enzyme. This enzyme is gained by cloning of penicillin acylase gene with recombinant DNA technique. For further study of recombinant DNA technique, it was carried out of the cloning of this gene by using E.coli B 130 as penicillin acylase resource, pFX-3 plasmid as vector, and E.coli JM109 as host cells. The transformants as ligation products from 410 ng pFX-3 (12 white colonies and 1 blue colony), were characterized by recombinant DNA plasmid analysis. Then the activity of penicillin acylase enzyme was tested microbiologically by using Serratia marcescens, there were 3 colonies which had penicillin acylase enzyme. text |
