Used fluconazole in the long term lead to many cases of fluconazole-resistant Candida albicans. To enhance the antifungal activity of fluconazole due to the case of resistance is by metal complex formation between antibiotics with metal or metalloantibiotics, such as with Cu (II), Ni (II), and Zn (II). These metal complex formation between antibiotics with metal reported can increased the efficacy of these antibiotics. The aim of the present study was to made metal complex of fluconazole with Mn (II) and Fe (III) as an effort to improve the biological activity of fluconazole. In this research, the manufacture of metal complex fluconazole with Mn (II) and Fe (III) using reflux method. Complexes formed then characterized using TLC, electrothermal, FTIR, DSC, TGA, and PXRD. Biological activity of the complexes was determined by antifungal potency test using disk diffusion method, 5+1 design. The activities of the complexes were compared with the ligand, fluconazole. In the TLC results, obtained that the complex Mn (II) and Fe (III) with fluconazole is pure and has similar polarity with fluconazole. Electrothermal analysis results showed that complexes Mn (II)-fluconazole and Fe (III)-fluconazole has a higher melting point than fluconazole. Fluconazole, complex of Mn (II)-flukonazol, and complex of Fe (III)-flukonazol had a melting point in a row from 136,4- 137,2, >250, and >170 °C. The results of FTIR analysis showed that the characteristic peaks of fluconazole at 1100-1630 cm -1 (phenyl group, aromatic C=C and C=N, and aromatic C-F) are shift between physical mixture of Mn (II) and Fe (III)-fluconazole with Mn (II) and Fe (III)-fluconazole complexes. DSC analysis results showed the thermal characteristics of fluconazole is different with the complexes. Fluconazole had an endothermic peak at 138,7 °C and an exothermic peak at 292,8 °C. Physical mixture of Mn (II)-fluconazole had two endothermic peak at 63,3 and 117,7 °C, and one exothermic peak at 218,6 °C. Mn (II)- flukonazol complex had a new endothermic peak at temperature 50,2 °C. Physical mixture of Fe (III)-fluconazole had two endothermic peak at 64,6 dan 253,3 °C. Fe (III)-flukonazol complex had three new endothermic peak at 75, 120-130, and 170-180 °C. PXRD analysis results showed that the Mn (II) and Fe (III) had different crystalline peaks compared with fluconazole. Mn (II)-flukonazol complex had characteristic peaks at pada 2?: 6,5, 7,5, 14, and 27,5° and Fe (III)-fluconazole complex had characteristic peaks at 2?: 11, 18, 22,5 and 25°. The complexes have been screened for their biological activity against Candida albicans. The results showed that Mn (II)-fluconazole has antifungal potency 3,57 fold higher compared with fluconazole and Fe (III)-fluconazole has antifungal potency 2,52 fold higher compared with fluconazole. Based on experiment result, Mn (II) and Fe (III)-fluconazole complexes can be made used reflux method with methanol as dissolving agent. These complexe has higher biological activity toward Candida albicans compared with fluconazole, 3,57 fold higher for Mn (II)-fluconazole complex and 2,52 fold higher for Fe (III)- fluconazole complex. text |
