The objectives of this research were to determine antimicrobial activity which represented with minimum inhibitory concentration and minimum bactericidal concentration of fungi isolated from Kuala Tanjung Beach North Sumatera against Staphylococcus aureus, Bacillus subtillis and Escherichia coli by using CLSI method (Clinical Laboratory and Standard Institute). Fungi isolates which had been isolated in previous study and had antibacterial activity, inoculated and purified on the YPD agar medium which consists of yeast extract, peptone and dextrose with ratio 5:5:10 on artificial sea-water. Purified fungi fermented on liquid YPD medium for three weeks. The result of fermentation filtrated to separate liquid medium with fungi mycelium. Liquid medium extracted by liquid-liquid extraction while mycelium extracted by maceration for 24 hours with ethyl acetate solvent. The extract concentrated and tested its antibacterial activity against Staphylococcus aureus, Bacillus subtillis and Escherichia coli with disk-diffusion method. The extract which had activity on concentration 250 µg/disk then being determined its minimum inhibitory concentration by using microdilution method. After that, minimum bactericidal concentration being determined at Mueller-Hinton agar medium. The result from activity test of four fungi isolated from Kuala Tanjung Beach, North Sumatera against Staphylococcus aureus, Bacillus subtillis and Escherichia coli, the range of inhibition zone diameter is 0,61 -1,8 cm. Five extract that showed antibacterial activity in 250 µg/disk (KT2 LLE extract against Escherichia coli; KT4 LLE, KT7 LLE and KT7 maceration extract against Bacillus subtilis; KT2 LLE against Staphylococcus aureus) have MIC values above 1024 µg/mL, while the greatest antibacterial activity was showed by KT2 maceration extract against Bacillus subtilis with MIC values 16 µg/mL and MBC values 1024 µg/mL. text
