develop resistance to some antibiotics, which can affect the therapy outcome. Other than
multidrug resistance, another important health issue is cancer, which is one of most
developed and deathliest disease in the world. In cancer therapy, one of the drugs’
mechanism is inhibition of topoisomerase I. One of the plant group that have shown
antimicrobaterial and anticancer potential is the Combretum genus. It is the largest genus
of Combretaceae. The genus comprises 370 species which distributed mainly in Africa and
Asia. C. latifolium, C. punctatum, and C. quadrangulare are types of Combretum plant that
exist in Indonesia, and there is no research done yet on their respective antimicrobial and
anticancer properties. This study aim to evaluate the antimicrobial and anti-topoisomerase I
activity of Combretaceae plants, C. latifolium, C. punctatum, dan C. quadrangulare.
Leaves of plant sample extracted by maceration with ethanol. Antimicrobial activity was
determined by disk diffusion method against Escherichia coli, Bacillus subtilis, and
Candida albicans. The lowest concentration which still provide antimicriobial activity then
used on microdilution method to determine its minimum inhibition concentration (MIC)
and minimum bactericidal concentration (MBC). To check its anti-topoisomerase I
activity, the extracts was then analyzed with Yeast mutant bioassay using well-diffusion
method against yeast-mutant Saccharomyces cereviciae strain 1140 which lacks of
Topoisomerase II but has Topoisomerase I. C. latifolium showed activity against E. coli
(MIC > 1.5 mg/mL), B. subtilis (MIC 1.5 mg/mL), and showed low inhibitory activity
against C. albicans. It did not shows anti-topoisomerase I activity. C. punctatum showed
low activity against E. coli, B. subtilis (MIC 1.5 mg/mL). It did not show activity against
C. albicans and as anti-topoisomerase I agent. C. quadrangulare showed activity against E.
coli (MIC 0.75 mg/mL and MBC 1.5 mg/mL), B. subtilis (MIC 0.375 mg/mL), C. albicans
(MIC 0.375 mg/mL) and has activity as anti-topoisomerase I agent. Ethanol extract of C.
latifolium and C. punctatum has low activity against Gram positive bacteria, Gram
negative bacteria, fungi, and has no activity as anti-topoisomerase I. Ethanol extract of C.
quadrangulare has strong activity against Gram positive bacteria, Gram negative bacteria,
fungi, and has activity as anti-topoisomerase I.
