Summary:Paracetamol was used as an analgesic-antipireutic agent, sold freely and could be obtained easily in single dose or combination with other active ingredients. Counterfeit drug was often defined with identity including the presence and amount of active ingredients are not appropriate to the compendial or spesification. This research was aimed to develop suitable analysis methode for paracetamol in tablet by determining the presence and amount of active ingredien for it applying to investigate a counterfeit drug. Paracetamol was extracted using ethanol then diluted until fix concentrate. Paracetamol solution was spotted on TLC plate then developed using a mixture of dichloromethane-methanol (8,5:1,5) as a mobile phase. Paracetamol spot was measured using spectrophotodensitometry at wavelength 247 nm. The analytical methods was validated. Calibration curve resulted a linear with of equation y = 7611.5x + 1604.7 and correlation coefficient (r) 0.999; limit of detection was 0.085 µg; limit of quantitation was 0.25 µg. Accuracy test of paracetamol standard showed with recovery of 100.308% 101.198%, 99.098% at 0.4; 0.8; 1.2 µg / spot and 98,712%, 100,692%, 101,995% for tablet formulation. Interday precision testing resulted with the coefficient variation (CV) of 1.464%, 1.1%, 1.443% and 1.849%, 1.29%, 1.1% for intraday precision. The method was validated and used for sample in the market. The presence of paracetamol was positive in all samples and the level of paracetamol were in the range of 96.18% - 107.72%.
