Cucurbita moschata which known as pumpkin is a plant that is widespread in Indonesia and contains many active compounds that could be used to prevent hyperlipidemia. This research aims to study the antihyperlipidemia activity of pumpkin seed. Pumpkin seeds are previously dried, grinding into powder form, then characterized. The experimental design consists of test for curative and preventive effects. The test was perfomed in female Wistar rat. For curative study, the rats are induce to have hyperlipidemia conditition by giving high cholesterol, propylthiouracil, and pure cholesterol for test of cholesterol and food high in carbohydrate for test of triglyceride. Two doses of pumpkin seed powder that are tested to prevent and cure hyperlipidemia are 1,35 and 3,15 g/kg body weight of rat. Rat lipid serum component which being determined are total cholesterol, HDL, LDL cholesterol, and triglycerides measured by using spectrophotometer after reacted with enzymatic reagent. Curative test for cholesterol has shown both doses of pumpkin seed powder tested can significantly (p<0,05) decrease total cholesterol level (204,39±32,64 mg/dL;163,35 ± 6,43 mg/dL vs. control: 305,63 ± 40,19 mg/dL), LDL cholesterol (72,40±16,12 mg/dL ; 48,16 ±12,29 mg/dL vs. control: 123,24± 15,38 mg/dL), and increse HDL cholesterol level (57,75±5,60 mg/dL; 86,53±31,54 mg/dL vs. control: 20,90±3,03 mg/dL) significantly (p<0.05). In curative test for triglyceride, both doses of pumpkin seed powder can decrease triglyceride (104,28±38,21 mg/dL ;133,57±30,35 mg/dL vs. control: 195,67±23,36 mg/dL) significantly (p<0,05). In preventive test for cholesterol both doses can prevent increase of total cholesterol (136,7±26,37 mg/dL; 95,55±11,78 mg/dL vs. control: 183,57±11,55 mg/dL), LDL cholesterol (81,63±17,46 mg/dL; 70,26±7,88 mg/dL vs. control: 119,71±15,24 mg/dL), and increase HDL cholesterol (58,30±11,34 mg/dL; 72,44±20,49 mg/dL vs. control: 20,94±3,42 mg/dL) significantly (p<0,05), whereas in preventive test for triglyceride, both doses of pumpkin seed powder can significantly (p<0,05) prevent increase of triglyceride (118±48,41 mg/dL; 95,28±27,87 mg/dL vs. control: 191±17,06 mg/dL). Based on the results, it can be concluded that both pumpkin seed doses being tested which are equivalent to 15 and 35 g powder used by human, which are given for 17 days in hypercholesterolemic and 14 days in hypertriglyceridemic, can decrease and prevent the increase of cholesterol total, LDL cholesterol, triglyceride level and increase HDL cholesterol significantly. text
