Summary: | Diarrhea is one of infectious diseases that kills the largest amount of human being.
Dysentery is diarrhea caused by infection of bacterial and is typically characterized by the
presence of blood or mucus in the stool. Bacterial that cause dysentery is Shigella, because
the caused is bacterial so for the treatment must use antibiotics. The patients of dysentery,
who live in the country known for the herbal medicines, often use antibiotics in same time
with herbal medicines as supplements or traditional medicine to enhance effect. The
objective of this research was to examine the antibacterial interaction between extract of
Curcuma xanthorrhiza Roxb, Allium sativum L., Sonchus arvensis L. with tetracycline as
antidysentery. Interaction activity antibacterial was conducted by in vitro and in vivo. On
in vitro study, activity antibacterial of the extract and the interaction with tetracycline was
tested by determination of the minimum inhibitory concentration (MIC) with microdilution
method, and minimum bactericidal concentration (MBC) with checker board method. On
in vivo study, the interaction of combination between tetracycline with the extract of
Curcuma xanthorrhiza Roxb or extract of Sonchus arvensis L. were conducted by
calculation of total plate count (TPC) bacteria in the feces of rats that had induced by
Shigella flexneri, the feces was cultured on the selective media (Macconkey Agar) for 8
days. The MIC of tetracycline, the ethanol extract of Curcuma xanthorrhiza Roxb, ethanol
and aqueous extracts of Sonchus arvensis L. respectively was 4, 128, 2048, 512 and 1024
?g/mL. The interaction of combination test plants with tetracyline showed synergistic
interaction. On the result of in vivo, at the 7
th
day of the treatment it was gived the result of
the combination tetracycline with ethanol extract of Curcuma xanthorrhiza Roxb, ethanol
and aqueous extracts of Sonchus arvensis L. had significant differences compared to the
positive control. From the research can be concluded that the interaction of tetracycline
with test plants had a synergistic effect against Shigella flexneri by in vitro study. From the
in vivo study, it showed that the combination of tetracycline with ethanol extracts of
Curcuma xanthorrhiza Roxb, ethanol extract of Sonchus arvensis L. and aqueous extract of
Sonchus arvensis L. could increase the antibacterial effect of the tetracycline.
