Summary:Piper betle and Piper crocatum are common plants in Indonesia. Both plants are widely used to treat an infection, swelling, and cancer. Due to that, it is alleged that both plants have immunomodulatory activity. This research investigates immunomodulatory activity of Piper betle and Piper crocatum leaf water extract. This research starts with collecting and characterizing plants, extracting active compounds with water, and the immunomodulatory test in BALB/c mice. Mice were divided into control group, Piper betle leaf extract at doses 21.5 and 43 mg/kg bw, Piper crocatum leaf extract at doses 32.5 and 65 mg/kg bw, and standard group (Zymosan A 10 mg/kg bw and methylprednisolone 15 mg/kg bw). Zymosan A and methylprednisolone were given by intraperitoneal injection while extracts through per oral route. Effect on nonspecific immune response was determined by carbon clearance test and organ index. Effect on specific immune response is determined through antibody titre. Piper crocatum aqueous extract at doses of 32.5 and 65 mg/kg bw showed immunostimulation effect with phagocytic index > 1,2. Piper betle aqueous extract at dose 43 mg/kg bw and Piper crocatum aqueous extract at dose 65 mg/kg bw induced humoral immunity indicated through primary and secondary antibodies titre which were higher than control. Based on this research, Piper crocatum water extract doses 32.5 and 65 mg/kg bw and Piper betle water extract at dose of 43 mg/kg bw can be used as immunostimulants.
