Summary:Rat is one of animal which has uricase enzyme. This enzyme will change uric acid as the end of product of purin breakdown becomes a soluble form, that is allantoin. So, naturally rat cannot undergo hyperuricemia. At this time, the study related hyperuricemia always use rats and intraperitoneal potassium oxonate as uricase inhibitor to induce hyperuricemia. Since potassium oxonate as uricase inhibitor, so it will inhibit changing process of uric acid becomes allantoin. In this study, hyperuricemia model was done by giving high purin foods which people with hyperuricemia or gout will avoid those kind of foods. This study is aimed to develop hyperuricemia model induced by high purin foods, that is melinjo seeds. This study used male Sprague Dawley rats. This study divided into two parts; 1) comparison ingredients of hyperuricemia induction to show the difference among induction method, 2) determination of optimum time to give antihyperuricemia substance to be tested. In first study, rats divided into 5 groups (n=3), those were normal group which given CMCNa 1%, group induction used melinjo seed suspension in 1% CMCNa (4.5 g/kg bw/day), group induction used chicken liver juice 20 mL/kg bw/day, group induction with mixed of melinjo seed and chicken liver juice (20 mL + 4.5 g)/kg bw/day, and group induction used ethanol extract of melinjo seeds 0.30 g/kg bw/day. Each ingredient was given for seven days. Bloods were taken from tail vena and examined by using Easy Touch ® GCU Blood Uric Acid and kit Uric acid-Liquizyme Uricase-PAP (Single Reagent). Rats in each groups were examined its uric acid by using those two methods before induction and checked its homogeneity. Then, uric acid examination was done in each two hour, four hour, and six hour after induction in H1 until H7 by using Easy Touch ® GCU Blood Uric Acid. The first study shows groups of melinjo seeds suspension undergo uric acid increasing after three days of induction and the average uric acid increasing can be observed both in two hour and four hour after induction. The next study was done by dividing rats into two groups (n=4); those were normal group which given CMCNa 1% and induction group which given melinjo seed suspension 4.5 g/kg bw/day. Rat bloods were collected from tail vena in H0, H3, H6, and H10 every before induction (T0) and three hour after induction (T3). Blood was examined by using kit Uric acid-Liquizyme Uricase-PAP (Single Reagent). Statistical calculation by using Mann Whitney U-Test to compare ingredients of hyperuricemia induction shows no difference between each of induction groups and normal group (p>0.05 and p>0.1). Statistical calculation by using independent t-test in the second study shows significant difference after 9 days of induction (p<0.05), meanwhile for statistical calculation in determining induction time by using dependent ttest to show difference between T0 and T3 and by using independent t-test to show difference of average difference of T0 and T3 between induction group and normal group show no difference in 0 th day, 3 rd day, 6 th day and 10 th day of induction (p>0.1 and p>0.05). Melinjo seeds can be used as ingredients to induce hyperuricemia based on the study have done for 9 days by giving 4.5 g/kg bw/day. This hyperuricemia rat model can be correlated that clinically melinjo seeds consumption in long term of time can cause hyperuricemia in human, because administration of melinjo seeds on rats with uricase enzyme can cause hyperuricemia.
