Summary: | Depression is a kind of mental disorders characterized by sadness, anxiety and loss of
interest or pleasure. It has been progressively increasing becoming a global public health
concern in the world. Synthetic drug used to treat depression affects the central nervous
system with a probable side effect of dependence, causing serious complication. Previous
reseach has shown antidepressant effect of nutmeg extract from its shell. This research was
aimed to further experiment active isolate of nutmeg that contributed most effect as an
antidepressant and dependence drug to be used as an alternative herbal medicine to treat
depression. Seven isolate was administered to seven groups, which are named A, B, C, D,
E, F and G with dose of 10 mg/kg. Positive control group was administered amitriptyline
and negative control group which was administered corn oil. All 9 groups were
administered drug, corn oil or isolate for 14 days straight. At day 15, all admistrations of
drugs was stopped for 24 hours. At day 1 and day 15, activity test which are open-field test
and forced swim test was commenced and analyzed. Group A shown most active result
from activity test was executed, its brains were dissected and used for ELISA analysis to
calculate noradrenaline and dopamine level. In open-field test, group A showed significant
difference (p< 0.05) in time spent in border on day 1 with value of 60.76±19.64 seconds
compared to negative group with value of 89.31±12.01 seconds. In forced swim test,
immobility time was measured and group A shown significant results (p< 0.05) on day 1
and day 15 with value 137.2±19.64 and 125.89±53.89 seconds respectively compared to
negative group with value on day 1 and day 15 of 201.76±17.89 and 197.91±21.57 seconds
respectively. Group A was chosen for ELISA analysis procedure. Group A shown
noradrenaline value of 1.42±0.01 ng/mL sample shows significant difference (p< 0.05)
compared to negative group. Group A also shows significant difference (p< 0.05) with
value of 1.37±0.07 ng/mL. Experiment shows isolate A from nutmeg’s (Myristica fragrans
Houtt.) middle shell with dose of 10 mg/kg mice which had been administered for 14 days
straight indicates antidepressant effect and dependence with anxiolytic effect.
