Alpha-amylase plays a role in the carbohydrate metabolism that breakdown long-chain carbohydrates such as starch into simpler molecules. The majority of Indonesian use medicinal plant as traditional medicine. Plants used as reduction of blood glucose level are crude drug bark Tinospora crispa L. (tinospora cordifolia), seeds of Leucaena Leucaena leucocephala (Lam.) de Wit (lamtoro), Momordica charantia L. (bitter gourd), Persia americana Mill. (avocado), Trigonella foenoem - graecum L. This study aimed to test the ability of alpha-amylase activity inhibition by compared reaction among alpha-amylase enzyme and substrate with reaction among alpha-amylase enzyme and extract. The result of the reaction is reducing glucose that can be measured by colourimetri after its reaction with 3,5-dinitrosalicylic acid. Its absorbances were measured by spectrophotomethry UV-Vis at wavelength 540 nm. Inhibition value were stated as percent inhibition using acarbose as positive control. Percent inhibition extract of tinospora cordifolia is 36.38±1.48%, lamtoro is 26.21±3.90%, bitter gourd is 49.63±4.12%, avocado is 52.88±2.02%, fenugreek is 20.67±6.17%, mahogany is 9.90±2.67%, sambiloto is 49.83±1.08%, jackfruit is 77.34±6.51%, nimba is 85.38±2.75%, pacing is 9.22±6.69%, mala is 74.78±4.10%, bodi is 19.63±6.16%, bungur is 12.07±3.47%, cecendet is 43.10±1.79%, angsana is 39.96±6.18%, golden purple is 22.03±6.02%, yacon is 3.22±1.44%, flower moon is 73.60±3.97%, patikan kebo is 45.87±8.09%, pomegranate 57.62±7.24%, bidara upas is 40.59±6.88%, acarbose 98.72±1.30%.
