DEVELOPMENT OF ANIMAL MODEL OF VERTIGO INDUCED BY STREPTOMYCIN AND GENTAMICIN IN FEMALE SWISS WEBSTER MICE

Vertigo is a hallucination of movement involving oneself (subjective vertigo) or the surrounding environment (objective vertigo) 1 . Bilateral vestibulopathy is one form of vertigo 2 . It is caused by chemical agents that are toxic to the sensory epithelium of the vestibular labyrinth, these m...

Bilateral vestibulopathy is one form of vertigo 2 . It is caused by chemical agents that are toxic to the sensory epithelium of the vestibular labyrinth, these medications include the common aminoglycoside antibiotics streptomycin and gentamicin 3 . The development of drugs for vertigo treatment need an animal model for preclinical testing. So, this research is aim to develop an animal model of vertigo that induced by aminoglycoside antibiotics, there are streptomycin and gentamicin. This study used female Swiss Webster mice which divided into 3 groups: negative control group, group with induced by streptomycin sulfate 260 mg/kg, and group with induced by gentamicin sulfate 70 mg/kg in two divided doses for 7 days. In the open field test methods, group induced by streptomycin sulfate showed significant decrease in mileage and increase duration in the border area on day 1 and 7, while duration in the central area of the box was declined on day 7, compared to normal conditions (p<0.05). Group induced by gentamicin sulfate showed significant decrease in distance and duration in the central area of the box, as well as increase duration is in the border area box open field on day 1 and 7 compared to normal conditions (p<0.05). In beam walking test, group induced by streptomycin sulfate did not show significant increase duration both on day 1 and 7 compared to normal conditions (p>0.05), and showed a significant reduction in the ability of mice passed wooden beams on day 1 compared to normal conditions (p<0.05), as well as showed a marked increase in the number of foot slips on day 1 compared to normal conditions (p<0.05). While groups induced by gentamicin sulfate showed no significant increase in duration and amount of slip on day 1 and 7 compared to normal conditions (p>0.05), and showed no significant decrease in the ability of mice passed the wooden beams on day 1 and 7 compared to normal conditions (p>0.05). In the forced swimming test, groups induced by streptomycin showed no significant increase on day 1 and 7 compared to normal conditions (p>0.05), while the gentamicin sulfate group showed no significant decrease on day 1 and no significant increase on day 7 compared to normal conditions (p>0.05). So antibiotic streptomycin sulfate 260 mg/kg and gentamicin sulfate 70 mg/kg in two divided doses for 7 days can be used to induce vertigo directed by indications of increased anxiety and balance disorder. text
