In this research, solid lipid nanoparticle (SLN) formulation containing synthetic Seramida was developed and later incorporated into gel and cream dosage forms. SLN can form occlusive system on skin surface to overcome Transepidermal Water Loss (TEWL). Method: SLNs were generated by high speed homogenization using ultrathurax and probe ultrasonicator. Experiment was started with lipids (stearic acid, glycerol monostearate, and apifil) and surfactants (tegocare and planta care) screening. SLN was characterized including to particle size analysis, polydispersity index, morphology (TEM), chemical and physical stability. Seramida SLN was then incorporated into gel or cream dosage form. Physical stability of the both dosage form were evaluated including to organoleptic, pH, and viscosity in room temperature and 40 0 C. Skin moisturizing test was later conducted using CM 825 corneometer. Result: Stable Seramida SLN was obtained with loading capacity of 4% of Seramida, 4% of apifil and 1% of plantacare as emulsifier agent. Particle size and polydispersity index of optimum SLN was 181.4 ± 3.20 nm and 0,306 ± 0.01. In period of 2 months, Seramida was relatively stable in SLN with final concentration of 92.26 %. Physical stability test showed that Seramida SLN loaded gel provided better stability than loaded cream. Water binding activity test revealed SLN Seramida was efective to maintain skin moisture. In addition SLN Seramida gel showed better water binding activity compared to blank gel and SLN Seramida cream was similar activity with blank cream. Conclusion: SLN Seramida was success prepared and physically or chemically relatively stable. Physical stability test showed that Seramida SLN loaded gel dosage form provided better stability than cream. Skin moisturizing test revealed that SLN Seramida loaded gel better than blanko gel and SLN Seramida loaded cream. text |
