Background and aim : Type 1 hypersensitivity reactions (allergy) occur as a result of repeated exposure to
antigen reaction is called an allergen where these allergens react with the IgE bound to mast cells and
basophil cells so that both release active mediators include histamine. Roots of eccinachea (Echinachea
purpurea L. Moench ) and herbs of meniran (Phyllanthus niruri L.) have immunostimulant effect, where
immunostimulant may enhance the immune response including antibodies. This study aims to assess the
effect of immunostimulatory preparations (extract Phyllanthus niruri and Echinacea purpurea) against type I
hypersensitivity in allergic male guinea pig by measuring wide of bump and reddish intensity parameters.
Method: Effect of immunostimulatory of the test preparation (Echinaceae extract 7.75 mg / kg bw; meniran
extract 3.875 mg / kg bw) of the active cutaneous anafilactive reaction on the back of allergic guinea pig
which was induced using egg whites and given orally (once daily) and again in days 1-14 and days 8-14 (for
allergic guinea pig ). Sensitized of allergic guinea pigs by egg whites 25% intraperitoneally (ip.) at first day.
In allergic of guinea pigs active cutaneous anafilactive reaction test was done by using challenges of egg
whites 2.5% and 5% by intracutan (ic.) on day 8. 12.15 was calculated from sensitization egg whites 25%
(H1). Active cutaneous anafilactive reaction was evaluated based on red bump happened and reddish
intensity. The data were processed statistically compared to control which were not given immunostimulant.
Results: A single dose of physiologist NaCl did not cause reddish bumps in guinea pigs. A single dose of egg
whites 25% in ip. can cause allergies in guinea pig on H8 (broad of reddish bumps was significant difference
in minutes120 challenged with egg whites 2.5% and the relative intensity of the redness in minutes 1440,
while challenged with egg whites 5% in minutes 30 and 1440 for intensity relative redness). Allergic
reactions on H8 for allergic control group wich was challenged by egg white 2.5% smaller than test 1 and test
3 and intensity relative redness at minute 30 to minute 120 was the same. Broad of reddish bump’s test group

intensity in minutes 30 (16,5%). Broad of reddish bumps on H12 for allergic control group wich was
challenged by egg white 5% smaller than test 1, test 2 (a significant difference in the minutes to 1440) and
test 3; relative reddish intensity was significantly differences with test 2 (33,33%), 3 (25,09%) in
minutes1440. Reddish bumps of test 2 is larger than all the other test (significant differences between test 1
and 2 in minutes 1440; test 2 and 3 in minutes 1440; test 2 and 4 in minutes 1440); the relative reddish
intensity was significantly between test 1 and 2 as well as test 1 and 4 in minutes to 1440). Broad of reddish
bumps on H15 for allergic control group wich was challenged by egg white 2.5% smaller than test 1, 2
(significant differences in minutes 60 ( 55,62%)); test 3, test 4; the relative reddish intensity in minutes 30-
120 for the test was greater than allergic control group, (except in minutes 120 and significantly differences in
the minutes to 1440 for test 2 (25,09%). Broad of reddish bumps of test 2 is larger than all other test followed
by a test group 3(significant differences occurred between test 1 and test 2 in minutes 60; test 2 and 3 in
minutes 60 and 90; test 2 and 4 in minutes 60 and 90; test 3 and 4 in minutes 90; and the relative reddish
intensity between test 2 and 3 as well as test 2 and 4). Broad of reddish bumps on H15 for allergic control
group wich was challenged by egg white 5% was smaller than test 2; test 3; test 4; relative reddish intensity
the test was larger allergic control (significant differences on test 3 in minutes test 60-120 (37,45% and test 2
in minutes 30-60 (28,32%) 1440 (28,32%)). Relative reddish intensity was significant differences between
test 2 and 4 as well as test 3 and 4 in minutes 30-120). Reddish bumps of test 2 is larger than all the other
tests group (significant differences occurred between test 1 and 2 in minutes 120 and 1440; test 2 and 3 in
minutes 120 and 1440; test 1 and 4 in minutes 60; test 2 and 4 in minutes 60-120; test 3 and 4 in minutes 90).
Conclusion: A single dose of egg whites 2.5% in ip. can cause allergies in guinea pig on H8. Echinaceae
root’s extract preparations when was given on H1-14, 8-14 and meniran herb’s extract wsa given at 1-14
could increase active cutaneous anafilactive reaction. In contras meniran herb’s extract could decrease that
reaction. The strongest increasement of that reaction was given by Echinacea root’s extract on H12-14 .
