Summary:Background and Purpose: The utilization of traditional herbal medicines has increased in society due to its nature origin that are considered safe. However, chemicals adulteration in herbal medicine is also increasing. One of the claims of traditional medicine revealed that it contains compound for increasing stamina. Addition of chemicals in traditional medicine with stamina turned out quite rapidly from year 2005-2013. It was not only containing sildenafil citrate, but also caffeine, paracetamol, tadalafil, and vardenafil HCl in a single dosage form or a combination. Therefore, it is necessary to develop a method of simultaneous identification of caffeine, paracetamol, sildenafil citrate, tadalafil and vardenafil HCl and tadalafil in herbal medicine preparation. Method: Method was begun with liquid-liquid extraction and continued with Thin Layer Chromatography (TLC) – Densitometry of CAMAG TLC-Scanner 4. The method used silica gel 60 F254 as the stationary phase. Mobile Phase for TLC was etil asetat, n-propanol, NH3 25% (45:5:1). Result: Rf values of caffeine, paracetamol, sildenafil citrate, tadalafil and vardenafil HCl and tadalafil were 0.57, 0.69, 0.27, 0.77 and 0.42, respectively. Resolution value (R) of caffeine-paracetamol, paracetamol-tadalafil, sildenafil citrate-vardenafil HCl, and vardenafil HCl-caffeine was obtained in a value of 2.0, 1.5, 2.4 and 2.4, respectively. This method have limits of detection (LOD) of reference standard for caffeine, paracetamol, sildenafil citrate, tadalafil and vardenafil HCl were 0.1475, 0.2273, 0.1346, 0.1951 and 0.2047 ?g(spot) ?1 whereas LOD of spiking were 0.3350, 0.3817, 0.1716, 0.3031 and 0.2659 ?g(gspot) ?1 , respectively. The method has applied to 9 samples traditional herbal medicines from the market. Result of the sampel analysis showed a mixture of caffeine, paracetamol and sildenafil citrate (A, B, and E), sildenafil citrate (F, G dan H), a mixture of paracetamol and sildenafil citrate (I), a mixture of caffeine and sildenafil citrate (C ), and tadalafil (D).
