The incidence and prevalence of gout have a direct relationship with the concentration of serum uric acid. Anredera cordifolia and Sonchus arvensis can be easily obtained and very easy to grow either in the lowlands or in the highlands. Previous studies showed that each of these plants has potential antihyperuricemic and inhibitory activity of the xanthine oxidase enzyme, so it is necessary to investigate simultaneously the inhibition of xanthine oxidase in vitro were confirmed by in vivo antihyperuricemic of each plant and its combinations of both. This study aimed to evaluate the inhibitory activity of the xanthine oxidase and the ability to reduce serum uric acid levels of 70% ethanol extracts of Anredera cordifolia and Sonchus arvensis leaves, and its combinations. Methods: Crude drug was extracted by reflux method using 70% ethanol. Inhibitory activity of the xanthine oxidase was determined in vitro by measuring uric acid formation by UV spectrophotometry. Antihyperuricemic activity was conducted in vivo in male Wistar rats that induced by high-purine diet and potassium oxonate. The reduction of serum uric acid levels after extracts administration was observed and compared to allopurinol. Results: The IC50 of Anredera cordifolia extract, Sonchus arvensis extract, and combination of both extract with a ratio of 1:1 were respectively 635.25 ppm, 1345.93 ppm, and 846.32 ppm. The IC50 of allopurinol as reference was 0.88 ?g/mL. Inhibitory activity of the combination extract against xanthine oxidase enzyme was additive. The results of in vivo assay showed antihyperuricemic activity of allopurinol, Anredera cordifolia extracts, Sonchus arvensis extracts, and the combination were significantly different compared to the positive control (untreated group) at 120 and 150 minutes after potassium oxonate induction (p<0.05). Combination extracts provided an additive effect in lowering serum uric acid levels in male Wistar rats. Conclusion: Extracts of Anredera cordifolia and Sonchus arvensis leaves have xanthine oxidase inhibitory and antihyperuricemic activities, and its combination provided an additive effect. The results were suggested that the extracts may have a considerable potential for developing as antihyperuricemia agent. text
