Description
Summary:Background and Purpose : Binahong is used as a potent anti-inflammatory medicinal plants on hematoma, hepatoprotective, relaxant on the gastric mucosa, antiobesity, hypolipidemia, improvement in renal function. Previous study showed that ethanol 96 % extract of binahong leaves improved renal function in renal failure rat model. Preliminary study showed that ethanol 70 % extract of binahong leaves gave larger decrease in creatinine and urea serum level compared to ethanol 96 % extract and water extract of binahong leaves. This study aimed to observe the influence of ethanol 70% extract of binahong leaves on serum creatinine, serum urea, the enzyme catalase, and organ histopathology index as parameters of renal function improvement. Method : Effect of ethanol extract in renal failure rat model was conducted by inducing nephrotoxic with combination of gentamicin and piroxicam. Parameters which were observed in this method were serum creatinine, serum urea, catalase enzyme, evaluation of renal function using renal index and renal histology. They were divided 5 groups: negative control group, positive control group, ethanol 70% extract with dose of 50 mg/kg bw/day, ethanol extract 100 mg/kg bw/day and the ethanol extract 150 mg/kg bw/day. All groups of test animals except the negative control group were induced with gentamicin (100 mg/kg bw/day) by intraperitoneally and piroxicam (3.6 mg /kg bw / day) orally for 7 days and subsequent oral administration of piroxicam to week 4. Improvement of renal function was seen after sample was given for 4 weeks of therapy. Results : The result showed that ethanol 70% extract of the binahong leaves with dose of 50, 100, and 150 mg / kg bw could repair the damaged function that was indicated by decreasing levels of serum creatinine, serum urea, enzyme catalase, and improvement in renal function index were significantly different with positive control (p<0.05). Based on observations of histology was showed that improvement in capsule Bowman and gromerulus by ethanol extract of binahong. Conclusion : Ethanol 70% extract of the binahong leaves with doses of 50, 100 and 150 mg / kg bw had effect in improvement of renal function in terms of serum creatinine, serum urea, enzymes catalase and renal index. Ethanol 70% extract of the binahong leaves with dose of 100 mg/kg showed better significantly improvement in renal function compared to positive control (p<0.05).
