Summary:Obesity and diabetic type 2 are public health issues which become world’s attention. Increasing prevalent of diabetic shows parallel pattern to obesity. Some expert called this as dual epidemic diabesity. Increasing of body mass index (BMI) and length of abdominal circumference are significant factors to obesity and diabetic type 2. Early research show that sambiloto and jati belanda respectively possess antidiabetic and antiobesity activity. Aim of this research was to investigate the activity of sambiloto and jati belanda extracts in treating diabetic conditions followed obesity. There are tho stages of induction. First stage is 42 days of high carbohydrate intake followed by insulin and glucose tolerance test and second test is intravena administration of aloxan 50 mg/kg bw. The animals were divided into 8 groups (negative control, positive control, metformin 195 mg/kg wb, glibenclamide 0.65 mg/kg bw, sambiloto extract 2 g/kg bw, jati belanda extract 0.5 mg/kg bw, combination of sambiloto extract 1 g/kg bw and jati belanda extract 0.25 mg/kg bw, combination of sambiloto extract 2 g/kg bw and jati belanda extract 0.5 mg/kg bw) and the extracts were given in 14 days. Parameters measured are blood glucose level and body weight. As preliminary test, glucose tolerance test was run on healthy animals and obesity induction method was chosen. In glucose tolerance test to healthy animals, each group showed the ability to suppress blood glucose level after glucose solution intake. As in glucose tolerance test for obese animals, sambiloto extract 2 g/kg bw, jati belanda extract 0.5 g/kg bw, and combination of sambiloto extract 2 g/kg bw with jati belanda extract 0.5 g/kg bw, can suppress the increasing of glucose blood level and reduce the rate of blood glucose level after glucose solution intake. After aloxan induction, combination of sambiloto extract 2 g/kg bw and jati belanda extract 0.5 g/kg bw was able to reduce both glucose blood level and body weight. As a conclusion, sambiloto extract 2 g/kg bw, jati belanda extract 0.5 g/kg bw, and combination of sambiloto extract 2 g/kg bw and 0.5 g/kg bw can treat prediabetic conditions caused by obesity while combination of sambiloto extract 2 g/kg bw and jati belanda extract 0.5 g/kg bw has the best antidiabetic activity to treat diabetic conditions with obesity compare to single use of sambiloto, jati belanda or combination of sambiloto 1 g/kg bw and 0.25 g/kg bw.
