INSTITUT TEKNOLOGI BANDUNG https://digilib.itb.ac.id/gdl/view/45982 Background and objectives: Radish and yam bean are known to reduce blood glucose level of several previous studies. Radish is a foodstuffs that prescribed as antidiabetic. Yam bean contains fiber and inulin to maintain normal blood glucose level. The objectives of this study to evaluate the effects of radish extract, yam bean extract and combination of them to reduce blood glucose level and increase insulin sensitivity. Methods: This study used two methods. The first method was injection of 55 mg/kg bb alloxan intravenously at a single dose. The mice were divided into seven groups: normal control, diabetic control, gliklazid 5,2 mg/kg bb, radish 200 mg/kg bb, yam bean 100 mg/kg bb, combination with high dose (radish 200 mg/kg bb and yam bean 100 mg/kg bb), combination with low dose (radish 100 mg/kg bb and yam bean 50 mg/kg bb). The mice were treated for 21 days. Blood glucose level was determined every 7 days and the ?-cell pancreas was observed by Gomori staining. The second method was aimed to insulin resistence by high fat emulsion 0,01 ml/g bb by intragastric for 21 days. The mice were divided into seven groups. Metformin 195 mg/kg bb as a comparator. Insulin sensitivity was determine using insulin tolerance method. Result: Radish 200 mg/kg bb, combination of high dose (radish 200 mg/kg bb and yam bean 100 mg/kg bb) and low dose (radish 100 mg/kg bb and yam bean 50 mg/kg bb) could reduced blood glucose level 45,50%, 34,33%, 50,22% were significantly in mice induced alloxan than diabetic control (p<0,05). In mice induced by high fat emulsion, single use of radish and yam bean could increase KITT value 125% and 191,25% were significantly than KITT value previously (p<0,05). Conclusion: Combination of high dose (radish 200 mg/kg bb and yam bean 100 mg/kg bb) and low dose (radish 100 mg/kg bb and yam bean 50 mg/kg bb) has an antidiabetic effect on damage of pancreas. Radish 200 mg/kg bb and yam bean 100 mg/kg bb can increase sensitivity of insulin hormone. text
