The richness of the ecological aspect and the biodiversity gave birth to a wide opportunity for novel natural organic coumpound exploration, include new compounds. Especially from the marine organism that nowadays has massively being studied for lead coumpound discovery. Of these marine species, sponge and symbiont microbes give the top contribution for new coumpound exploration. Sponge have developed a unique metabolism adaptation due to their lack of the motoric organ evolution, of which is in their chemo- and symbio- metabolic adaptation. Microbial communities live densely in the sponge tissue and correlate each other forming a total metabolism for the sponge adaptation. This unique adaptation had been a key of success for sponge well being in critical condition, not only modification in the sponge organism physiology, but also occur on the symbiont that produce a unique biogenesis. Endophytic Fungi in sponge tissue is a symbiont that provides a wide opportunity for lead compound exploration. In this research, a sampling of marine sponge is due at Kepulauan Seribu National Park, a sponge identified as Clathria reinwartii has been isolated and an endophytic fungi had been obtained, identified through BLAST method as Mucor circineloides. The fungi was cultured in 3 kg rice media and 19 grams of ethyl acetate extract of the culture has been seperated with chromatography purification methods (Gravity Column Chromatography, Vaccum Column Chromatography, Radial Chromatography, and Medium Performance Liquid Chromatography. metabolites have been isolited, compound was characterisized with 1H NMR, 13C NMR, HSQC, and HMBC measurement, the structure was established as polycketides and a benzofurans derivatives: AR1 (bis-2-ethylhexyl-terephtealate), AR2, AR3, and AR4-AR5. The compounds were tested with in-vitro P388 Murine
Leukemia bioactivity test and respectively shows IC50 value at: 72,64 Mg/mL; 17,77 Mg/mL; 19,26 Mg/mL; & 6,76 ?g/mL. The phytochemical study in Mucor circinelloides from Clathria reinwardti in Kepulauan Seribu National Park coastline is in the very first time to be researched. |
