Summary: | Rheumatism is a disease that attacks the joints and surrounding areas which includes
osteoarthritis, rheumatoid arthrtis and gout. Gout or often called uric acid disease is rheumatic
disease that affects many people. Gout is a condition where there is an increase in uric acid levels
above normal, causing a buildup of uric acid. Pirdot (Saurauia bracteosa DC.) and remason
(Polygala paniculata L.) are plants that people traditionally use to treat gout. Tests were carried
out to prove the potential of pirdot and remason to treat gout by testing the inhibitory activity of
xanthine oxidase ethanol extract and water extract of both plants. The inhibition of xanthine
oxidase activity was measured based on the level of uric acid formed in the reaction of the
xanthine substrate with the xanthine oxidase enzyme, compared to the reaction of xanthine
substrate, xanthine oxidase enzyme and addition of sample then measured by UV visible
spectrophotometer at 290 nm wavelength. The test results showed that remason ethanol extract
had the highest inhibitory activity of xanthine oxidase with IC??at 504,84 ± 5,06 µg/ml. While IC??
of pirdot ethanol extract was obtained at 533,64 ± 4,37 µg/ml, IC??of pirdot water extract was
1.341,87 ± 14,70 µg/ml and IC??of remason water extract was 1.060,71 ± 9,57 µg/ml. Remason
ethanol extract which had the highest activity was fractionated using n-hexane, ethyl acetate and
water solvent and then tested the xanthine oxidase inhibition activity. Ethyl acetate fraction of
remason showed the highest activity with IC??at 175,65 ± 3,11 µg/ml. The results of
phytochemical screening and TLC showed that remason ethanol extract and remason ethyl
acetate fraction contains flavonoids which can provide xanthine oxidase inhibitory activity.
