Summary:Immunomodulators are compounds that can regulate the immune system which can be obtained by synthetic or isolated from plants. This research aims is to develop an immunomodulator fromf the plant. Two plants were selected to be studied in this research i.e. ciplukan leave and vetiver root. Active compounds from ciplukan leaves and vetiver roots were extracted by boiled in water for 30 minutes. Immunomodulatory activity of extracts of both plants is determined by using carbon clearance method, organ index, antibody titer, and cytokine levels determination. Immunomodulatory effect of the extracts was demonstrated in animal models of Adjuvant Induced Arthritis (AIA) obtained by intraplantar injection of Completes Freund's Adjuvant (CFA). Effect against rat rheumatoid arthritis model is determined by measured the legs volume, joints thickness, arthritis score, and TNF-?level. Through immunomodulatory effect test, water extract of ciplukan leave (EADC) at doses of 20 and 60 mg/kg bw and water extract of vetiver root (EAAW) at dose of 15 mg/kg bw showed immunosuppressive activity with phagocytic index value <1. In addition, both exracts did not increase the formation of the primary and secondary antibodies titers. Both extracts decreased IFN-?level, however not significant compared to than that control. These data indicated that, at doses tested those extracts have immunosuppressant activity. Effects of those doses of the extract on animal models of rheumatoid arthritis showed a decreased in arthritis scores (p<0.05) significantly. Highest inhibition of legs swollen and joints thickness are occur after administered i.e., 51.1 and 21.0%, respectively EADC at a dose of 20 mg/kg bw. After 21 days of extract administration, both extracts at those doses tested decreased TNF-?level, however only EAAW at a dose of 15 mg/kg bw decreased TNF-?significantly (p<0.05) compared to control. Through micropathology examination, EADC at a dose of 20 mg/kg bw and EAAW at a dose of 5 mg/kg bw could repair hyperplasia synovial membrane.
