Description
Summary:
Abstract:

A research on the isolation, selection and bioassay of lignin degrading microorganisms has been carried out in two steps. The aim of this experiment was to obtain lignin degrading microorganisms. The first step was microbial isolation from pulp waste water, using nutrient agar medium for bacteria and potato dextrose agar medium for fungi. The second step was to select the isolates based on the capability of using lignin as carbon source in basal mineral medium plus lignin. The selected isolates was identified and them found out their optimum growth conditions such as pH of medium, incubation temperature and waste concentration. Those optimum conditions were used in lignin degradation process. There were 15 isolates and two among them namely Pseudomonas a/cal/genes and P. stutzeri were able to degrade lignin compound. These two isolates were used as inoculum for lignin degradation process with the concentration variation (%v/v) of 0%, 5%, 10%, 15% dan 20% and the incubation time were 7, 14 and 21 days. The observed parameters were lignin residual concentration, COD and BOD. The result showed that the decreas in lignin concentration, COD and BOD during 21 days incubation by P. alcaligenes were 59.12%, 72.06% and 58.69% while by P. stutzeri were 40.57%, 78.3% and 46.86%.
