Summary:Hypertension is a chronic disease characterized by an increase in systolic blood pressure > 140 mmHg and diastolic blood pressure > 90 mmHg. Tempuyung (Sonchus arvensis (L.)) is a plant has been widely known by Indonesian people as a medicine, and it is used empirically in the treatment of various disease for example is hypertension. This study aims to determine the antihypertensive effect of tempuyung leaves with approach in vivo and in vitro method. Study antihypertensive effect of tempuyung leaves use extracts water and ethanol dosage forms tested. Testing antihypertensive effect in vivo was performed using non invasive measuring blood method on male Wistar rats using instrument Kent Scientific's CODA system and the induction of hypertension using epinephrin 0.5 mg / kg bw. A test dose of water extract are 50, 100, and 200mg kg bw, ethanol extract are 50, 100 and 200 mg/kg bw, and propranolol 0.9mg / kg bw as reference drug. Test results show the water extract dose of 50 and 200mg/kg bw and the ethanol extract doses 100, and 200 mg/kg bw able to inhibit the increase diastolic and systolic blood pressure if compare with the positive control group (p <0.05). The test results also showed water extract dose 50 mg/kg bw and ethanol extract 100 mg/kg bw showed similar pharmacological effect when compared to propranolol 0.9 mg/kg bw (p> 0.05). Testing antihypertensive effect in vitro carried out by the enzymatic reaction, the test aims to determine IC50 water extract and ethanol extract, and captopril is used as standard to inhibit activity of ACE. The test results showed IC50 water extract and ethanol extract of Sonchus arevensis (L.) leaves to inhibit ACE activity ware 50,8 ?g//mL and 46,71 ?g//mL and captopril IC50 1.26 ?g//mL. The study results showed that the tempuyung leaves have potential as antihypertensive with the mechanism of inhibiting an increase systolic and diastolic blood pressure with induced by epinephrine on test animals and also able to inhibit the activity of ACE.
