Summary:Nanofiber has been developed for a variety of medical applications, for example in healing therapies for bacterial infected wounds. This study aims to determine the effect of two different types of emulsifier (a combination of polysorbate 80 and span 80, and lecithin) on the characteristics, release profile, and antimicrobial activity of PVA / short chain chitosan (COS) which is added by eugenol as an active substance. Nanofiber is made through the electrospinning method with fixed parameters in the form of a flow rate of 40µL / min, an electrical voltage of 15kV, and a distance between the tip of the needle and the collector 12cm. Concentrations of PVA and COS are optimized and optimum PVA results of 10% and COS of 5% are obtained. Nanofiber PVA / COS-Em / Eu and PVA / COS-Lc / Eu are formed with average fiber diameter sizes of 280 ± 126nm and 224 ± 79nm, respectively. The results of morphological observations with SEM nanofiber PVA / COS-Em / Eu are in the form of interlocking fibers and large beads, while the PVA / COS-Lc / Eu nanofibers are long smooth fibers. The presence of eugenol in both nanofibers was confirmed by observing the FTIR spectrum. The results of the evaluation of release showed that eugenol from PVA / COS-Em / Eu nanofiber had a faster release process than the PVA / COS-Lc / Eu nanofiber. Even so, the results of the evaluation of antimicrobial activity against Staphylococcus aureus showed the same good activity.
