In the case of investigation of toxic effect from tramadol, its content in biological samples needs to be confirmed. This study was aimed to model detection method in subjects exposed to adulterated alcoholic beverage containing tramadol by qualitative detection in hair sample of mice. Pharmacological effect of the adulterated beverage was confirmed by observing analgesic activity using tail-flick method. The mice were induced 4 times within 22 days of experiment. They were divided into four groups, each received 3.63 mg/kg tramadol, 1.81 mg/kg tramadol, 3.63 mg/kg tramadol mixed with 0.7 ml/kg ethanol, and 1.81 mg/kg tramadol mixed with 0.7 ml/kg ethanol, respectively. The results showed inhibition of analgesia in all test groups which was significantly different (p<0.05) from control (given only Sodium-CMC solution). Qualitative analysis on hair sample using Fouriertransform Infrared Spectroscopy (FTIR) showed aromatic moiety at peak 1650 indicating tramadol in groups treated with tramadol and ethanol. Taken together, the results suggest that hair sample can be used to detect tramadol for its confirmation in the case of toxicity from adulterated alcoholic beverage containing tramadol. text
