Indonesia’s tropical ocean is rich in microalgae. whose diversity needs to be studied because microalgae are called cryptic species, where the morphology looks the same but can be distinguished genetically. Genetic identification is an effective technique that can be used to differentiate cryptic species. The purpose of this study is to analyze inorphologically and genetically microalgae originating from marine areas in the Part Island, Indonesia. Methodology used in this study includes sampling of sea water conteining microalgae, screening of microalgae single cells, growth the cell isolates to obtain a pure culture, cells observation under light microscopy, frustule observation under SEM (scanning electron microscopy), isolation of total DNA, PC’R amplification and sequencingof the l8S rDNA V4, r6‹’L-3P and LSU D2 / D3 marker ge nes, nucleotide sequence analysis using DNA baser assembler V5, Bioedit, BLAST (NCBI) and MEGA 7 for building phylo6enetic trees. The results showed that 3 isolates of single cell microalgae were successfully screened and cultivated. Two of them, namely isolates PsTE dan CPK were successfully analyzed morphologically and genetically. while one other isolate, GsTE, was only morphologically analyzed. The cell morphology of the GsTE isolate showed the cell shape of pennate cell (elongated; bent at the edge), the cell size of about 70 pm and cell color of brownish yellow color. This morphology das similarities with the genus Gyrosi gma. The cell morphology of the PsTE isolates showed the cell shape of the pennate cell (elongated), the cell length was around 63.7 pm. and the cell color of brownish yellow. This morphology has similarities with the genus t’leuro.Sigma. Thi s result was confirmed by the genetic data, which showed 98.78% similarity to /’/eurosigiria 7nreroiediiim (NCBI: AY485489. 1) based on 18S rDNA V4, and 9ß.47°.é similarity to I’leurosi gma sy. (NCBI : KX981822. 1) based on rbcV-3P. Hence the PsTE isolate was identified as Pleuros ignio up. PsTE. Cell morphology of CPK isolates showed similarity to the t"haeio‹'eros genus, that was a centric cell shape, that forming chains and having .sefoe. The cell was about 3.7 pin in cell length with a greenish-yellow in color. This result is confirmed by genetic data, showed 99.43% similarity to f ’hael‹ ‹’ero.s fester.s.s*mits (NCBI: MG9l4624. 1) based on D2/D3 LSU gene. Hence the CPK isolaie was identified as I ’haeioceros tenuissirrius CPK. text |
