To obtain maximum results, lipase needs a specific environmental condition, such as pH and temperature. This study investigated the effect of concentration and type of buffer on Plumeria sp. latex lipase activity. Enzyme activity analyzes with products hydrolysis percentage measurement through acid value analysis. The results of this study show the highest lipase activity gain by ammonia salmiak buffer with 0,5 M concentration. Lipase activity on 0,5 M buffer concentration is 79,153%. This value is very good because the average of maximum lipase activity is 80%. This percentage shows the amount of triglyceride which successfully hydrolyze into fatty acid. Different type of buffer used could affect lipase activity. Chaotropic type of buffer should be avoided because could inhibit lipase. Chaotropic characteristic of buffer could be observed from Hofmeister series. The fatty acid product could be used as resources of another process. By optimizing the lipolysis reaction condition, a large amount of fatty acid could be obtained using a cheaper method. One of the processes which using fatty acid as the resource is metal soap production. With the results of this study, it is expected that the production of fatty acids for making metal soap will produce more products with an easier method. text
