Summary:The combination of paracetamol and caffeine has a function as an analgesic and antipyretic. The combination is mostly available in the form of a tablet. This study aims to develop a validation method for analysis of paracetamol and caffeine concentration using UV spectrophotometry with multivariate calibration, namely Partial Least Square. PLS was developed using 25 training sets and 9 test sets. Paracetamol and caffeine concentrations were used from 4 - 20 ppm and 2.5 - 12.5 ppm, respectively. The analysis was carried out at a wavelength of 220-300 nm at 1 nm intervals and then processed with PLS using the Minitab 17 program. Then validated by correlation coefficient (R2 ), Root Mean Square Error of Calibration (RMSEC), Root Mean Square Error of Cross-Validation (RMSECV), % recovery and Root Mean Square Error of Prediction (RMSEP). In this research, the R2 value obtained for paracetamol and caffeine were 1. The RMSEC of paracetamol and caffeine were 0.0199 and 0.0197. The RMSECV of paracetamol and caffeine were 0.087 and 0.086. The RMSEP of paracetamol and caffeine were 0. For the accuracy, the % recovery of paracetamol and caffeine were 100%. The developed method has been validated and can be applied to combination paracetamol and caffeine.
