Rice bran is a plant-based lipase source alternative that can be easily found in Indonesia. Before use, lipase from plants needs to be prepared first to remove water and oil while maintaining lipase in an immobilized state in a dense portion of oil-free grains without losing its lipolysis activity and even, if possible, increasing it. Lipase powder preparation is done by soaking rice bran in organic solvent. The solvent is expected to be able to dissolve oil in the seeds, keep enzymes from denaturation, and increase the stability and activity of enzymes. This study is aimed to obtain a procedure for preparing rice bran lipase powder which provides the best activity and can be used repeatedly to hydrolyze oil. Variants of the solvents used in this study were acetone, cyclohexane, a mixture of acetone and diethyl ether, a saturated solution of glycerol and calcium formate in acetone, a mixture of 1 : 2 v/v cyclohexane-acetone, and a mixture of 1 : 2 v/v cyclohexane-saturated solution of glycerol and calcium formate in acetone. Lipase powder activity test was carried out by lipolysis reaction for 5 hours. Lipase powder activity was calculated from the acid number of lipolysis products. Based on the results, the most suitable procedure to prepare rice bran lipase powder is to use only acetone as the solvent because it produced the highest activity (0.56%) compared to other solvent variations. text
