Previously it was found that cocrystal formed between sodium diclofenac and L-proline (NDP) which showed increase in solubility. In this study, it was further known that in that cocrystal there were four hydrates in the crystal lattice which were confirmed through thermal analysis using thermogravimetry (TG). In general, the presence of hydrates can reduce solubility so that some efforts are made to reduce the amount of hydrate in cocrystal. By heating the NDP tetrahydrate cocrystal using an oven as well as by the formation of cocrystal in minimal water condition, it was known that another hydrate form of NDP cocrystal was formed. Characterization by thermal analysis with TG, with Fourier-transform infrared spectroscopy (FTIR) and diffractometry with powder X-ray diffractometry (PXRD) showed the formation of NDP monohydrate cocrystal and differences between NDP monohydrate and tetrahydrate. The solubility of NDP monohydrate and tetrahydrate showed increases compared to sodium diclofenac in both water and pH 6.8. Moisture stability test showed that after one hour of being exposed in a room with 75 ± 5% relative humidity, NDP monohydrate turned into NDP tetrahydrate. text |
