Summary:Phenylethyl Resorcinol is a synthetic compound derived from natural lightening compound, Scotch Pine Bark. It is regarded as a reliable whitening agent and widely used in bleaching cream. In the market, those products are usually added with other compounds such as preservative to inhibit the growth of microorganisms. In order to control and ensure the safety of products in market, an analytical method to determine Phenylethyl resorcinol is needed. The method to analysis should be specific and selective, so it can provide correct analytical signal in a mixture of analyte within sample, without any interaction between analyte. For this purpose, an analytical method using High Performance Liquid Chromatography (HPLC), UV detector 282 nm, Zorbax Eclipse Plus C18 ?m (4.6 x 150 mm) column was developed. Gradient system was used in mobile phase, at 1 - 4 minutes was used Acetonitrile-Water (30:70), at 4.1 - 15 minutes was used Acetonitrile-Methanol-Water (40:20:40), and the flow rate was 1.0 mL/min. Validation parameters performed were specificity, linearity, limit of detection (LOD), limit of quantitative (LOQ), accuracy, precision, and robustness. The result of each parameters from analysis of Phenylethyl resorcinol with addition of five preservative matrix and other bleaching agents are, specificity gave resolution >1.5, linearity in range 10.48-73.36 mg/mL and the regression equation obtained was y=8175.30x-2709.61 with r = 0.9999 and Vx0 = 0.78, the percentage of recovery was 97.46 –103.05%, the intraday and interday precision was obtained HorRat value ?2, indicating the precision of analytical method was already fulfill the requirement. Limit of detection (LOD) value was 0.7183 mg/L and limit of quantification (LOQ) was 2.3943 mg/L. Those result showed that this analytical method is selective, linear, accurate, and precise. This method can be used for testing of Phenylethyl Resorcinol in skin whitening creams.
