Summary: | According to WHO (World Health Organization), the number of asthma patients in the world reached 339 million people. This number will increase as people’s lifestyle changes and air pollution increases. Salbutamol is one of the commercial asthma medicines commonly used. Consequently the optimization of salbutamol synthesis reaction is very important. In this study, we reported the journey throughout the synthesis of salycyl alcohol and 1-[3- (chloromethyl)-4-hydroxyphenyl]ethanone which are the precursor in salbutamol synthesis. Reaction between phenol and formaldehyde to obtain salycyl alcohol was unsuccessful and led to polymerization. Substituting the phenol with p-hydroxy acetophenone. 1-[3- (chloromethyl)-4-hydroxymetyl]ethanone is obtained (3) from the reaction between p- hydroxy acetophenone (1) with formaldehyde (2) in HCl 37%. The product was purified by column chromatography and characterized with 1H-NMR spectroscopy.
