In Indonesia, the traditional treatment for diabetes overcome many uses of plants . Plants that use are Cinnamomum Burmani Ness, ex. BI., Androraphidis paniculata (Burm.f.) Ness), Phaleria macrocarpa (Scheff) Buerl., Annona muricata L., Momordica charantia L., Mangifera indica L. (Bapang), Mangifera indica L. (arum manis), Myristica fragrans Houtt., Tinosporae crispa (L) Miers., Annacardium occidentale L., Vinca rosea L., Ocimum bacillicum L., Orthosiphon stamineus Benth., Garcinia mangostana L., Annona squomosa L., Psidium guajava L., Zingiberis officinale Roscoe, Averrhoa bilimbi L., and Camellia sinensis (L.) O. Kuntze. include green tea, black tea, and white tea. Methods: The method used in this research is of ?-amylase inhibition activity (in vitro) by enzymatic analysis method Bernfeld (1955), Odhav (2010). Plants that have the highest inhibition activity has been isolated its active compound. Results: The ethanol dried extract Mangifera indica L. (91.74%) and Garcinia mangostana L. (91.7%) had inhibition ?-amylase activity are relatively higher then other plants. Ethanol extract of Mangifera indica L. ) were fractionated by liquidliquid extraction and continued testing of ?- amylase inhibition activity . Ethyl acetate fraction has ?- amylase inhibition activity higher than n- hexane fraction and water fraction. Ethyl acetate fraction was isolated active compounds and obtained two compounds. The compounds are characterized by UV - Vis spectrophotometry method, 1 HNMR and 13 CNMR, and the results can be concluded elucidation of these compounds are gallic methyl and mangiferin. IC50 ?–amylase inhibition activity ethanol extract of Mangifera indica (9,47 µg/mL), ethyl acetate fraction (19.69 µg/mL ), gallic methyl (243.11 µg/mL), mangiferin (91.49 µg/mL), and acarbose as a comparison (3.93 µg/mL). Conclusion: Dried ethanol extract of Mangifera indica L. have ?-amylase inhibition activity (91.74%) higher than other plants. Ethanol extract of Mangifera indica L. have IC50 values (9.47 µg/mL) were higher than ethyl acetate fraction and active compound.. Isolated active compound are gallic methyl (methyl 4,5,6 trihidroksibenzoat) and mangiferin (2-C-????-D-gluko-1,3,6,7-pyranosyl tetrahidroksixanton).
