High Performance Liquid Chromatography (HPLC) is the most effective methods for sugar analysis compared to other methods, because it has fast analysis time and simple sample preparation. For sugar analysis, amine group silica column is needed to get a good separation. In this study, 700-800 nm diameter Si-NH2 column was prepared from silica modified with 20% of N-[3-(Trimethoxysilyl)propyl]ethylendiamine (N-TMSPen. Synthesis of monodispersed silica was carried out using a Tetraethyl Orthosilicate (TEOS) as precursor, an isopropanol as solvent, a water as hydrolyzing agent and an ammonia as catalyst. Silica particles also characterized using Scanning Electron Microscopy (SEM) dan Fourier Transform Infrared (FTIR) Spectroscopy with KBr pellet method. Observations of the FTIR spectrum obtained some new peaks, such as peaks from - CH2 (3050-2980 cm-1)¸ Si-C (950-800 cm-1), C-N (1250-1020 cm-1) and primary N-H (1650- 1550 cm-1) which indicates that the Si-NH2 synthesized has been successfully done. Afterwards, the modified silica particles was loaded into three types empty HPLC columns, which are 150 mm x 4.6 mm, 100 mm x 4.6 mm and 50 mm x 4.6 mm with HPLC pump help and all of them provides a maximum pressure below 300 bar for 1 mL/minute flow rate. This pressure value indicates that the column created has the potential to be used in HPLC systems which can provide a maximum pressure of 600 bar. text
