In this research, the purification of xanthorrhizol from methanol extract of temulawak rhizome was carried out through two stages of chromatography, namely vacuum liquid chromatography and radial chromatography. The characterization of the molecular structure of xanthorrhizol was carried out using NMR spectroscopy. Furthermore, this research has also been carried out to optimize the separation of xanthorrhizol by the vacuum liquid chromatography method using CaO as a stationary phase. The parameter varied during optimization was the ratio of the extract mass to the mass of CaO from 1: 160 to 1:20, with n-hexane as an eluting solvent. The best optimization occurs in the ratio of extract mass to mass of CaO was 1:80 with the number of elution 8 times, each 30 mL. In this condition, the total mass percentage of the eluted extract was 67%, where the rich fraction with xanthorrhizol was in the 3rd to 8th fraction text |
