This study aimed to investigate the role of cyclooxygenase pathway in the pathology of ASD using mice model. ASD was modeled in 6-8 weeks old mice born to mother treated with 500 mg/kg bw sodium valproate (VPA) at gestational day of 12,5. The methods used were measurement of sniffing time on exposure to intruder mice and the measurement of COX activity in some areas of the brain. Behavioral observation was carried out in four groups of mice, namely normal, and mice model of ASD pretreated with normal saline and diclofenac at 1,2 and 3 mg/kg bw. The measurements of COX activity were carried out in frontal cortex, hippocampus, and cerebellum. The results showed a decrease in sniffing time in mice model of ASD, which was reversed by pretreatment with diclofenac. The average of sniffing time in normal mice was 154,33±9,88 sec, while that in mice model of ASD was 24,83±5,11 sec. Sniffing time in mice model of ASD pretreated with 1,2 and 3 mg/kg bw of diclofenac were 46±4,81 and 76,83±8,13 sec, respectively. Results of measurement of COX activity increased indicated the tendency of increases in activity in frontal cortex and cerebellum, while a decrease in hippocampus of the brain of mice model of ASD compared to normal mice. Overall, results of the present study suggest the role of cyclooxygenase pathway in the pathology of ASD. text |
