INSTITUT TEKNOLOGI BANDUNG https://digilib.itb.ac.id/gdl/view/50002 Nowadays, treatment of envenoming snakebites that use an antivenom which produce from Immunoglobulin-G (IgG) in mammalian serum has some disadvantage, such as animal ethics problem. Its considered violate animal right cause of routine blood draw from that animal. The alternative one for envenoming treatment is produce Immunoglobulin-Y based antivenom, from aves yolk, such as chicken. Compared with production from mammalian antivenom, IgY based antivenom has some advantages including ethics factor, because of antivenom could taken from egg yolk. Easiness of handling and maintenance chicken, especially in cost, and also large amounts of antibody production are other advantages of IgY based antivenom. However efficacy of IgY in body didn’t known yet. The purpose of this research to study distribution and elimination kinetics of Immunoglobulin-Y in blood stream. IgY isolated based on Pauly method (2011) then injected to dextra marginal ear vein of rabbit slowly. Then blood draw from sinistra central ear artery of rabbit at certain time. Concentration IgY in blood serum sample have been analyzed with Direct ELISA. The result showed that at first injection, concentration of IgY in blood serum sample increased rapidly from 10th minutes and reached steady state at 2 hours which indicated IgY distribution in blood flow occurred rapidly at rate 6,11/hour. IgY elimination observed after 24 hours that occurred slowly from blood flow cause of natural elimination system and immune system of rabbits body. IgY was undetected at 168 hours (first injection), 144 hours (second injection), and 120 hours (third injection). Consecutive elimination rate at first, second, and third injection are 0,23/hour; 0,16/hour; 0,23/hour. Based on paired t-test there aren’t shows significance results (p>0.05), that third of all didn’t have difference after 24 hours. The result also showed that blood draw from Vena Saphena in rabbit foot at 10th minutes was detected in ELISA. text
