Many therapies for nicotine dependence have been developed such as nicotine replacement therapy (NRT) and bupropion therapy, but still, there are side effects, including uneasiness, that can lead to therapy failure. This study was aimed to investigate the effect of 5-lipoxygenase (5-LOX) in nicotine dependence. Mice were given intraperitoneal nicotine at 0.5 mg/kgBW once daily for 5 days to induce dependence, and to determine the role of 5-LOX, oral zileuton (1; 3.2; and 10 mg/kgBW) were given to the assigned groups of mice, 30 minutes before nicotine injection. Dependence were evaluated with Conditioned Place Preference (CPP) protocol. The preliminary results showed that zileuton dose-dependently lower the preference score, indicating essential role of 5-LOX in the development of nicotine dependence. The brain substrate that involves 5-LOX in nicotine dependence was hypothesized from the result of reviewing several relevant references. text
