This study was aimed to investigate the effect of 5-lipoxygenase (5-LOX) in nicotine dependence. Mice were given intraperitoneal nicotine at 0.5 mg/kgBW once daily for 5 days to induce dependence, and to determine the role of 5-LOX, oral zileuton (1; 3.2; and 10 mg/kgBW) were given to the assigned groups of mice, 30 minutes before nicotine injection. Dependence were evaluated with Conditioned Place Preference (CPP) protocol. The preliminary results showed that zileuton dose-dependently lower the preference score, indicating essential role of 5-LOX in the development of nicotine dependence. The brain substrate that involves 5-LOX in nicotine dependence was hypothesized from the result of reviewing several relevant references. text |
