Summary:<b>Abstract:<p align=\"justify\"> <br /> It has been known that human mitochondrial DNA (mtDNA) is highly polimorphic due to its high mutation rate. D-loop is a region of human mitochondrial genome that has highest polimorphism. An analysis of the D-loop region nucleotide sequence can be used for human identification or origin of population because this mtDNA is maternally inherited without recombination with paternal genome and available in abundance.<p align=\"justify\"> <br /> Previous research has identified ten sequences of Indonesian D-loop nucleotide from ten ethnics and show a variance (morph). An observation about the D-loop nucleotide sequence of maternal lineage individuals is needed to know whether the variance is significant or not. Nucleotide sequence of mitochondrial genome D-loop region of Indonesians maternal lineage from 3 generation (grandmother, mother and daughter) have been determined in this research. Each mtDNA is amplified at its D-loop region with PCR technique using M1 and M2 primer. 0,4 kb fragment as a result of amplification is cloned with pGEM-T vector in JM109 E. coil cell.<p align=\"justify\"> <br /> The recombinant plasmid were isolated and then sequenced using Dideoxi Sanger method. The result of these sequencing show that 443 bp nucleotide sequence of mitochondrial genome D-loop region of Indonesians maternal lineage have 100% homology.
