Excessive levels of uric acid in the blood can cause hyperuricemia and gouty arthritis. In addition, the process of uric acid formation also causes the formation of superoxide anion radicals and hydrogen peroxide and cause an inflammatory reaction. Kelor, tempuyung, and sambiloto are plants that have traditionally been used as medicinal plants in Indonesia. Kelor leaves contain some compounds such as quercetin, isorhamnetin, apigenin, and chlorogenic acid which have the potential effect as xanthine oxidase inhibitors. Tempuyung leaves contain kaempferol, quercetin, and myricetin that have the potential as xanthine oxidase inhibitors. Sambiloto herbs contain andrographolide and flavonoid compounds such as apigenin and luteolin which have potential as xanthine oxidase inhibitors. But until now, there has been no research on the isolation of active compounds from plants which also involves testing the inhibitory activity of xanthine oxidase from certain compounds in these three plants. This study was conducted to obtain botanical information and opportunities in the development of these materials as candidates for natural ingredients to reduce uric acid levels, and to determine the potential activity of ethanolic extract of kelor, tempuyung and sambiloto leaves as inhibitors of xanthine oxidase enzymes through in vitro test of xantin oxidase enzyme inhibition. Test results on the sample indicate that plants have inhibitory activity against xanthine oxidase with the greatest inhibition was found in the sample of sambiloto herbs ethanolic extract with a concentration of 12,5 ppm by 43,668% and the least inhibition percentage was found in sambiloto herbs ethanolic extract with a concentration of 62,5 ppm estimated at 15,223%. However, no correlation was found between the concentration of samples and percentage of inhibition. While allopurinol as a positive control showed a strong xanthine oxidase inhibitory activity, concentration that could inhibit activity by 50% (IC50 value) was obtained at a concentration of 6,134 ppm. text
