Summary:Staphylococcus aureus has ability to internalize cell with the complex intracell persistence mechanism. Due to this characteristic, the conventional way to deliver the antibiotic to target the infection site will be less effective. NLC structure modified with chitosan conjugated with acemannan is developed to be the antibiotic carrier and serve the purpose the enhance the cellular uptake of the antibiotic and eventually the higher treatment effectiveness will be achieved. NLC is prepared with solvent emulsification and evaporation method which has been optimized in the previous study. Characterization of the NLC product regarding the particle size, potential zeta, particle distribution size, and morphology should be conducted to ensure whether the product has met the qualification. Stability during storage within some periods of time can be maintained and improved when the product is freeze dried. Cryoprotectant such as sucrose and trehalose in the range of concentration within 5-15% should be added to protect the product during the process. Freezing and drying ?±??±?¬???± ð??±?ð?¬®®??­¬?®± ?ð?í ã?(glass transition) and Tc (collapse temperature) point, also the proper rate and duration of each process should be applied to obtain the optimum product. The freeze-dried NLC modified with chitosan-acemannan is characterized to determine its quality and to predict the stability during storage. In vitro assay with cell is conducted to determine the cell viability, ???­?®???¬­ð?ð????ð??±??¬?ð????± ?í± ®±??????¬?d the ability of product to kill the intracellular bacteria.
