Summary:Vannamei shrimp (Litopenaeus vannamei) is one of the most common shrimp species cultivated in Indonesia. Vannamei shrimp cultivation is a potential export commodity. The obstacle in shrimp farming is a disease caused by a virus. White spot syndrome virus (WSSV) is one of the most threatening diseases in the vannamei shrimp culture industry. DNA-based molecular testing is an accurate and precise method to detect virus infection. In the first step of identification process, the DNA will be multiplied through polymerase chain reaction (PCR). PCR is a technique that amplifies a DNA template to produce specific DNA fragments in vitro. PCR involves thermally cycling DNA at three temperatures. Each cycle doubles the amount of DNA. This project emphasizes the temperature control design of the DNA-based WSSV detection system in vannamei shrimp. The thermocylcer rely upon Peltier-effect thermoelectric heating and cooling. Peltier will heat the thermal block. The thermal block is where the substrate is placed during the amplification process. The thermal block temperature is controlled by PID controller. Heated lid is used to prevent condensation on the top of substrate. The heated lid is set to have higher temperature than substrate temperature. The control method used in the heated lid is Bang-Bang control. The controller is implemented using the ESP32.
