<b>Abstract:<p align=\"justify\"> <br /> <br /> An experiment to study the effect of elicitor derived from Verticillium dahliae Kleb. on gossypol content of Gossypium hirsutum L. cv. Tamcot root-culture has been conducted. Root-culture grew best on liquid Linsmaier & Skoog (LS) medium with addition of 10-6 M NAA (Naphthalene Acetic Acid). Root-culture was elicited with elicitor derived from autoclaved V. dahliae Kleb. The concentrations of elicitor used were 40, 60, and 80 gg DW/mL. The harvesting times were 2, 4, and 6 days after elicitation. The gossypol content was analyzed qualitatively and quantitatively by using High Performance Liquid Chromatography (HPLC). The highest content of gossypol in root-culture was obtained by addition of 80 gg DW/mL elicitor. The highest content of gossypol in root (4,427 ± 0,544 mg/g DW) was obtained on 2 days after elicitation, the percentage of increment was 38,39%. The highest content of gossypol in medium (11,610 # 0,791 mgt) was obtained on 4 days after elicitation, the percentage of increment was 62,81%.<p align=\"justify\">
