The freeze-thaw method is a hydrogel synthesis method that is able to form a stable structure and is not toxic to the human body. PVA is a material that has excellent biocompatibility. Meanwhile andrographolide (AND) is an active substance that has antibacterial activity and comes from the sambiloto plant (Andrographis paniculata). In this research, hydrogel-PVA synthesis has been carried out with various concentrations of the bioactive compound AND using the freeze-thaw method for wound dressing applications. The results obtained showed that the viscosity value of the solution decreased with increasing AND concentration. The results of pH measurements are in normal conditions 5,877–5,930 which has the potential to inhibit bacterial growth. The results of FTIR characterization are related to the XRD test results, namely that there is a molecular interaction between PVA and AND which causes a shift in the wave number in the O-H and C = O stretching regions, as well as the addition of amorphous doamin after synthesis in the form of a hydrogel. The results of SEM characterization and porosity test showed that the PVA / AND hydrogel had a porous structure with the size of the porosity increasing with increasing AND concentration. Which is related to the test results, the degree of development, and the increasing weight loss. Meanwhile, in contrast to the results of the density test, mechanical properties, melting heating value and the degree of crystallinity are decreasing. This is due to the encapsulation of AND which interferes with the hydroxyl group formation process in PVA, and the cross-linking between the PVA chains. This hydrogel with a porous structure is useful for helping the wound healing process because it is able to absorb wound exudate and help the encapsulation process of AND which has antibacterial properties. PVA/AND hydrogel antibacterial activity increased with increasing AND. PVA/AND-4 hydrogel had the highest antibacterial activity of 27.476%. Therefore, the PVA/AND hydrogel has the potential to be used as a wound dressing application. |
