The purposes of this study are to isolation khusimol compound from vetiver oil which will be used as marker and determine khusimol content at various drying times after harvesting. Methods: Fractionation of vetiver oil was performed by classical column chromatography using gradient elution. Subfractionation was conducted by classical column chromatography using isocratic elution. Purity test was carried out by single development TLC using three mobile phases with different polarity. Characterization and identification of isolate applying NMR spectroscopy. Determination of khusimol content in extract at various drying times was performed by TLC-densitometry. Results: Target compound was found in combination of fraction 3 (GF-3). Furthermore GF-3 was subfractionated by classical column chromatography. Purity test of combination of subfraction 3 (GS-3) by single development TLC using three mobile phases with different polarity, showed 1 spot. 1 H-NMR and 13 C-NMR data of isolate X as the same as 1 H-NMR and 13 C-NMR data of khusimol compound in literature. Molecular formula of khusimol C15H24O. Khusimol content in extract at various drying times after harvest time 0 h, 6 h, 12 h, 24 h, 36 h and 48 h were 0,00195 ± 0,00002; 0,00215 ± 0,00002; 0,00230 ± 0,00003; 0,00281 ± 0,00001; 0,00172 ± 0,00003 dan 0,00138 ± 0,00002 g/100 g. Conclusions: The odor contributor compound was khusimol (C15H24O). The highest khusimol content was revealed by vetiver extract which drying for 24 hours after harvest. Drying vetiver more than 24 h could reduce khusimol content. text |
