Description
Summary:Abstrak :

Land farming is a technique in bioremediation that uses soil as media This laboratory scale bioremediation research tried to investigate oil sludge biodegradation by using fertilizer (urea) as nitrogen source and sawdust as bulking agent. The experimental field was a box 25 x 10 x 15 cm3 that contained mixture of 2% sawdust and 2 kg native soil).. Mix culture with the same ratio of six hydrocarbonoclastic bacteria isolated from the crude oil sludge were used as inoculum after 3 times adaptations each in 0%, 3%, and 5% of crude oil sludge in Stone Mineral Salt Solution (SMSS) added by 0.01% yeast extract. Growth pattern that was observed by using fluorescein diacetate (FDA) probe showed that the highest enzyme activitiy of 2°d and 3`d adaptation occured in 72 hour (FDA hidrolized 65.53 µg/ml and, 68.73 pg/ml). Treatments in this research consisted of 12 kinds based on C : N ratio and with/without inoculum. Parameters examined were growth through enzym activity performance by using FDA, pH, and percentage of crude oil degradation by using gravimetry method where humidity maintained ± 60%. Treatments were stopped when level of crude oil degradation has reached more than 70%. Investigation showed that E2 after 40 days treatment had the best result with 99.9% crude oil degradation, FDA hydrolyzed 19.73 µg/ml, pH 6.49 ± 0.37, temperature 26.11 ± 0.7 °C, and humidity 58.73 ± 5.49 %. Next step was El and E2 analyzed by using Gas Chromatography (GC) to examine pattern of degradation. Analysis by GC showed that there was biodegradation of Unresolved Complex Mixture, such as polyaromatic, branch-cycloalkane, and branch - alkane. Compared with El, which had no inoculum addition, E2 was faster in biodegrading crude oil. These findings indicate that mix culture of isolated bacteria have good capability to degrade crude oil, where E2 is the best treatment for biodegradation of crude oil, and adaptation steps of inoculum is important to faster biodegradation 14,22% during treatment.
