Summary:<b>Abstract :</b><p align=\"justify\">Somatic embryogenesis is development of embryo or embryogenic tissues from somatic cells. Development of somatic embryo and zygotic embryo are almost similar in morphology and molecular features, therefore it can be used as a model to study plant embryogenesis. Up to now. initiation and development of somatic embryo are still debatable. For that reason, the present research is focused on somatic embryo development derived from cotyledon of mungbean (T-\'igna radiata (L.) Wilczek), with purpose to determine the initial cell of somatic embryo. Embrvogenic calli were induced from cotyledon of mungbean in MS medium supplemented with 5 mg/L 2,4-D. The calli were then subculture in MS medium supplemented with 0,5 mg/L NAA, 0,01 mg/L BA, 0.01 mg/L kinetin, 0,01 mg/L 2ip and 0,1 mg/L GA3. The calli were transferred to liquid L6 medium supplemented with 0,01 mg/L GA3. 0,1 mg/L picloram, 0,5 mg/L kinetin. 200 rng/L casein hydrolysate, and 2% sucrose. Histological observation on callus showed that several meristematic cells derived from cotyledonary parench_vm cells were form in or beneath the calif surface. These cells were potential to become embryogenic. On liquid medium, pro-embryo developed from round embryogenic cells. Three types of pro-embryo were found. namely pro-embryo without \"suspensor\", pro-embryo with \"suspensor\" and two pro-embryo on one \"suspensor\". These pro-embryo were subsequently developed into globular and heart shapes embryo. Based on the results of this experiment, we concluded that in liquid medium somatic embryo of mungbean cotyledon resulting from single round cell which actively underwent cell division to form embryo. Up to heart stage, developmental sequence of this somatic embryo was similar to zygotic embryo development.
